Identification of multiple components in deep eutectic solvent extract of Acanthopanax senticosus root by ultra-high-performance liquid chromatography with quadrupole orbitrap mass spectrometry

Acanthopanax senticosus is an important medicinal and edible homologous plant that is widely used in medicines, health products, and functional beverages. Phytochemicals present in plants are partly responsible for various health benefits. Traditional extraction methods use a large number of high-risk organic solvents (acetone, chloroform, etc.) that are potential threats to the environment, human health, and food safety. However, a persistent problem remains in this field with regards to extraction using a simple "green" method. Therefore, a deep eutectic solvent (DES) was used instead of traditional high-risk organic solvents to extract the phytochemicals from Acanthopanax senticosus root. In this study, DES was synthesized from choline chloride and lactic acid, which was used as a substitute for a strong organic solvent to extract saponins, phenylpropionic acid, and terpenoids from Acanthopanax senticosus root. The phytochemicals extracted from Acanthopanax senticosus were analyzed by ultra-high-performance liquid chromatography with quadrupole orbitrap mass spectrometry in positive and negative ionization modes. A total of 91 types of phytochemicals were identified from DES extracts and 17 chemical constituents were newly identified in the root of Acanthopanax senticosus via primary and secondary high-resolution mass spectrometry (MS1/MS2). Furthermore, DES was demonstrated as a promising functional component of the extraction solvent, and its non-toxic properties endow broad application prospects in the food and health products industries.
