The missing link!!! A carboxylic acid functionalized Meldrum's acid derivative (3-(2-methyl-4,6-dioxo-1,3-dioxan-2-yl)propanoic acid) was identified as a valuable building block to achieve facile late-stage functionalization of enzyme-responsive “covalent-assembly” probes whose activation produces fluorescent coumarin or pyronin dyes. The ability of such cyclic C-nucleophile to promote both in situ formation of a photoactive compound and release of a molecule of interest will facilitate the implementation of “covalent-assembly” principle to small-molecule based theranostics.
