To cycle or not to cycle: macrocycles play a fundamental role in various areas of chemistry, although their preparation is nontrivial. In this work, the synthesis of a highly substituted biomass-derived chiral pyrrolidine and the difficulties of “click” cyclization are reported. After a detailed DFT study, we propose a simple and useful parameter to anticipate the feasibility of related transformations.
