A standalone route for synthesis of BCX-1777 and BCX-4430: By application of an addition–reduction–cyclization synthetic strategy, an advanced intermediate was prepared from Boc-protected lactam and dihalogenated pyrrolopyrimidine, in a chemoselective and stereoselective manner. This strategy provided BCX-1777 and BCX-4430 in an improved overall yield from a common precursor.
