DNA Tiling Enables Precise Acylation-Based Labeling and Control of mRNA

A general method that enables the site-localized acylation of mRNA at virtually any determined sites is described. The TRAIL method utilizes a novel DNA tiling strategy by hybridizing a pool of inexpensive “protector” DNAs to block mRNA and combining with an “inducer” DNA to extrude a reactive RNA loop for acylation. This approach can be employed for both mRNA labeling and control for multiple applications.
