Rapid identification of high-affinity aptamers against human TNFα: In vitro selection coupled with microbead-assisted capillary electrophoresis (MACE) separation was performed to rapidly identify anti-TNFα aptamers; it was completed in only three rounds. Selected Apt14 showed the strongest inhibition activity according to the cell viability assay. Furthermore, by introducing a complementary linker to the aptamers, facile dimerization was performed, resulting in enhanced affinity and inhibition activity compared to that of the monomer.
