Analogues of Pam2Cys, the traditional TLR2 ligand, were designed to probe structure–activity relationships, namely, the individual importance of each ester carbonyl to TLR2 activity is called in to question. Analogues possess ethers in place of either one or other of the ester functional groups. Access to these analogues is provided by union of classical enantiomerically-enriched starting materials through the key S-alkylation of a protected cysteine.
