Completely Chemically Synthesized Long DNA Can be Transcribed in Human Cells

DNA segments of 78 bp and 258 bp in length, were prepared by chemical ligation of chemically synthesized DNA fragments with phosphorothioate and haloacyl terminus groups. The 258 bp DNA, coding shRNA for luciferase genes with a truncated SV promoter sequence, was successfully transcribed and showed full gene silencing effects in human cells.
