The catalytic residues in the cyclase domain of the class IV lanthipeptide synthetase SgbL were identified and validated in this study. It was demonstrated how each of these residues is crucial for the formation of the methyllanthionine β-thioether crosslinks from Cys thiols and dehydroalanine/dehydrobutyrine residues in the modified globisporin precursor peptide SgbA(S20T).
