The Effect of Acute Unilateral Denervation Injury on Purinergic Signaling in the Cholinergic Synapse

Previously, we found a change in the efficiency of the modulatory action of ATP under the influence of some non-physiological factors at neuromuscular synapses in rodents. This study was aimed at evaluating the effect of ATP on synaptic transmission of isolated rat soleus muscles after denervation injury. It was shown that rats exhibited an increase in the strength of electrical stimulation-evoked contractions of the soleus muscles after 7-day denervation. Application of ATP increased the electrically evoked contractions of the intact soleus muscle but had no effect on the denervated muscle. The nonselective P2-receptor antagonist suramin abolished the effect of ATP. We speculate that the reduction of ATP activity in the rat model of denervation is most likely caused by a decrease in the number of P2-receptors on the nerve terminal because of impaired anterograde transport and decreased nerve fiber conduction velocity.
