Single-molecule enzymology can be used to profile heterogeneous enzyme activity in clinical samples. A diluted sample is mixed with a substrate, and the mixture is loaded to a microwell array. The change in fluorescence over time in wells containing an enzyme is used to quantify the enzyme activity, resulting in a distinct fingerprint for the target enzyme. This fingerprint is used for the quantification of multiple enzyme isotypes in the sample.
