The ring system of purine is remarkably stable, but can be opened at either side, if properly activated. In this review, purine ring opening reactions leading to imidazole and pyrimidine derivatives are summarized. Purine ring opening is also a powerful tool for the synthesis of new ring-modified purine derivatives (e. g., N1-labelled analogs) by various rearrangement pathways.
