Using synthetic PEG-NAD(H) cofactor swing arms, we demonstrate the modular assembly of dehydrogenases to produce a series of novel, self-contained synthetic transhydrogenases. We also show that the stabilities and catalytic properties of the resulting transhydrogenases are improved without detriment to the selectivities of the native active sites.
