Analysis of the Effect of Vitamin C at IC50 on RAW264.7 and K562 Cells Based on 1H NMR Metabonomics

Daily use of vitamin C as a health care supplement is widespread, but toxic effects of excessive doses or long-term usage of vitamin C are not well studied. Here, we first determined the IC50 of vitamin C using RAW264 cells (920 mM) and K562 cells (800 mM) using an in vitro cytotoxic assay. Next, cell extracts were obtained 24 h after vitamin C treatment at IC50 and analyzed for differential metabolites by nuclear magnetic resonance (1H NMR) metabonomics using multivariate statistical analysis software. The results showed that there was a significant difference (p < 0.01) in cell growth between the vitamin C group and the control group in both RAW264.7 cells and K562 cells. We found that vitamin C treatment affected the galactose metabolism, methionine metabolism, inositol metabolism, glyceride metabolism, and glycine, serine, and threonine metabolism in both cell lines. This study provides a theoretical basis for exploring the toxic effects of high doses of vitamin C.
