Improving the pre-harvest sprouting resistance of rice cultivar IR36 using wild rice ( Oryza rufipogon ) W630

Pre-harvest sprouting is one of the most serious problems in rice production. Sensitive pre-harvest sprouting causes low seed quality and losses before harvesting. Seed dormancy is one of the most important traits relative to pre-harvest sprouting. In this study, the common wild rice, Oryza rufipogon W630 (strong seed dormancy) was used as a donor to improve the pre-harvest sprouting resistance of O. sativa Indica IR36 (popular cultivar in Asia). Thereafter, 170 backcross recombinant inbred lines (BRILs) at the BC2F8 generation were produced by the single seed descendant method. Their degree of dormancy was assessed and genotype was analyzed using 180 polymorphic SSR markers covering entire chromosomes. Finally, a QTL analysis was conducted by composite interval mapping to identify putative QTLs of seed dormancy and the wild allele effects to the background of typical rice cultivar IR36. Thus, two of 170 BRILs showed stronger seed dormancy compared to recurrent parent IR36. In addition, two putative QTLs for seed dormancy at chromosome 2 and 7 were also found in this population through composite interval mapping. These results indicated the stronger dormancy trait of two BRILs derived from wild rice Oryza rufipogon W630; this trait was also controlled by genetic factors.
