In vitro morphogenic responses in Centaurea depressa, Bieb

Centaurea depressa, a medicinal herb used in traditional Ladakhi medicine is classified as vulnerable under the IUCN Red data book. Reports on its in vitro micropropagation for conservation of this species is however lacking. This study showed that nodal explants multiplied (11.75 shoot buds/node) when BA (2.22 µM) was used while in KN lesser number of multiple shoots were induced. NAA even at the lowest concentrations (0.54µM) completely reduced this response and induced mostly calli. The most interesting result was the induction of shoots (5.2± 0.39 shoot buds induced/explant) from root/rhizomatous explants of this species incubated in light (16 h) and cultured in ½ MS basal medium with BA (4.44 µM). Anatomy of the regenerated vascular traces in the induced shoot confirmed direct organogenesis and is a useful method to propagate this endangered species. DNA phylogenetic analysis with universal barcoding primers (ITS1, rbcL, matK, psbA-trnfH and ycf1) using total DNA isolated from the tissue cultured propagules is presented.
