HPLC Assay of Rutin Content in the Leaves of the Black Poplar

A method for assay of rutin content in the leaves of the black poplar (Populus nigra L.) using HPLC at a wavelength of 360 nm was developed; this used an Elsico column (No. 10302, 2 mm × 80 mm; Silasorb C18 7 μm), with a mobile phase consisting of acetonitrile-water (2:8) supplemented with 1% glacial acetic acid. The suitability of the method for analysis of standard rutin samples was confirmed. Rutin content in black poplar leaves ranged from 0.67% to 1.05%. Statistical analysis of the experimental results provided evidence that the error level in single determinations of rutin in black poplar leaves with a 95% confidence interval was ± 4.6%.
