ROPs are molecular switches controlling diverse cellular and developmental processes. In this issue, Ge et al. (pp. 1484–1499) report that ROPs are targeted to the plasma membrane not only through the activity of RhoGDIs but also directly by vesicular trafficking, involving COPII, ARF1, AP‐1, and sterols. RhoGDIs and the vesicular trafficking route synergistically mediate ROP targeting and thus cell polarization.
