Linear Splines for Shelf Life Analysis of a Drug Product Stored in Hybrid Storage Conditions

Abstract

Large molecule pharmaceutical products frequently require refrigerated storage at 2–8 ¨C to maintain their efficacy and safety over the duration of their shelf life, but there are benefits if a product can be stored at room temperature, such as 25 ¨C, for the last few weeks of its shelf life, prior to use. With this type of “hybrid storage,” common methods for estimating shelf life may not be applicable because the products stability trends may change when the product is moved from refrigerated storage to room temperature storage. We propose using linear splines to extend a common method for estimating shelf life and accommodate the change in the stability trend. We show that the proposed method works well with a simulated dataset.
