We appreciate the continued interest in our work and the extensive and constructive discussion provided by David Parks in his comment. We slightly disagree with the notion that the increments in sensitivity (Q) and the lower detection limit (DL) of spectral cytometry as compared to conventional cytometry, which we have described, are not significant. In our opinion, they are not only of statistical but also of operational significance. The experiments proposed by David Parks, aiming at a multivariate evaluation in situations of practical relevance, are well designed to underpin this pragmatic significance. In our original publication (1), we had already pointed out that the calculation of Q, B and DL based on the added values of the individual detection channels ignores the vector properties of the multispectral measurement. To account for this, we had introduced a multivariate overlap factor (O).
