Increased efficiency: The combination of azide‐functionalized HIPS or tandem Knoevenagel ligation reagents and SPAAC enables the fast and efficient mono‐ and double‐derivatization of proteins with stoichiometric amounts of DBCO‐modified payloads at low concentration.
