Harnessing synthetic biology to enhance heterologous protein expression

Expressing heterologous proteins in microbial hosts is a core technology for protein science. However, heterologous protein expression often produces low yields and/or misfolded products. To overcome these challenges, a protein of interest (POI) can be joined to another protein that endows the fusion with favorable properties for expression and/or purification. All the currently used fusion partners are derived from natural sequences that evolved for other purposes. Here we demonstrate that novel proteins, which did not evolve in nature, can serve as fusion partners; and these De novo Expression Enhancer Proteins (DEEPs) facilitate high level expression and facile purification of a range of heterologous proteins and peptides.https://onlinelibrary.wiley.com/doi/10.1002/pro.3907
