Effect of UV-A on vincristine biosynthesis and related peroxidase isozyme changes in Catharanthus roseus

ABSTRACT

This study revealed effect of UV-A on the vincristine accumulation and peroxidase isozymes changes in the Catharanthus roseus plantlets and relationship between them. Effect of the UV-A on the C. roseus plantlets survivor was largely because the plantlets (two weeks age) died after irradiation by UV-A more than 30 minutes. The electrophoretic patterns of peroxidase isozymes extracted from the seedlings C. Roses of the different treated 0, 10, 20 and 30 minutes, showed the changes in numbers and intensity of peroxidase isozymes bands. Using HPLC analysis the vincristine concentration extracted from the plantlets of C. Roses which were treated by UV-A revealed that the increase in vincristine concentration values were 368, 411 and 431 µg/g dry plant leaves (DPL) corresponding 10, 20 and 30 minutes respectively, compared the control 0.0 time 331 µg/g (DPL). the UV-A can be used to stimulated the biosynthesis of the metabolite in general but, this stimulation was dependent on the irradiated time exposure.
