Fluorescence quantum yield determination using simultaneous double-beam absorption measurement

Fluorescence quantum yield determination using the Simultaneous Absorption and Fluorescence Emission (SAFE) method can be performed on the spectrofluorometers which are equipped with the transmittance detector. However, the commercial spectrofluorometers are not commonly equipped with the transmittance detector, and those which possess this functionality are offered only in the single beam geometry. This seriously limits the application of the SAFE method. This article describes a simple approach to adapt a sample compartment of virtually any spectrofluorometer model for SAFE quantum yield determination. Furthermore, it shows the first application of a double-beam absorption measurement for spectrofluorometers. This approach provides the utmost signal stability, is inexpensive, and requires no hardware programming.
