Metabolic engineering of Escherichia coli for the production of neryl acetate

Abstract Neryl acetate, a monoterpenoid derivate originated from neryl diphosphate (NPP), has found extensive applications in food, agriculture, and cosmetic fields. In this study, neryl acetate was biosynthesized in recombinant Escherichia coli for the first time. First, the alcohol acetyltransferase, ATF1, was heterologously expressed in a nerol-producing E. coli strain. The resultant engineered strain, LZ006, accumulated 0.250 ± 0.014 mg/L of neryl acetate. Further overexpression of tHMG1, which improved the metabolic flux of MVA pathway, enhanced the yield to 8.064 ± 0.724 mg/L in LZ007, representing a 32.2-fold increase compared to that of LZ006. Finally, supplementation with 2 g/L pyruvate in the culture further increased neryl acetate production to 11.712 ± 0.653 mg/L, representing a 46.8-fold increase over that of the LZ006 strain. This study provides a novel method for the biosynthesis of neryl acetate and the strategies described here could be adapted to biosynthesize many other NPP-derived bioproducts.
