A focused library of natural product‐inspired flavonoids was prepared through diversity‐oriented synthesis with systematic variation of chemical properties, scaffold, stereochemistry, and appendages. The library was evaluated to assess biological performance diversity and to identify chemical diversity elements that predict diverse biological activities.
