Keratinocytes produce cytokines and participate in immune responses and skin healing. Poly(I:C) is a ligand for TLR3 that induces the production of IL-8 in skin and promotes wound repair. Bee venom (BV) has many effects on skin, and Nakashima et al. show that BV synergizes with various TLR ligands to enhance the IL-8 production in HaCaT cells (a human keratinocyte cell line). This is especially marked for high-molecular-weight poly(I:C), with responses to TLR1/TLR2 and TLR2/TLR6 ligands also enhanced. BV-sPLA2 is the active component for this synergy not only in HaCaT cells but also in primary human epidermal keratinocytes and the enzyme activity is required.
