In a recent paper in Cell, Yao et al. used the Mosaic Analysis with Double Markers (MADM) genetic system for granule neuron precursor (GNP) cell-specific simultaneous ablation of the tumor suppressor Tp53 and the expression of fluorescent label in a few cells in a Ptch1-heterozygote, Tp53-heterozygote mouse to track these clonally related populations during development of medulloblastoma.1 Lineage tracing revealed that tumor-associated astrocytes, as well as medulloblastoma cells, are derived from GNPs. Transcriptome profiling performed by Yao et al. demonstrated that GNP-derived, tumor-associated astrocytes clustered closer to normal astrocytes than GNP-derived tumor cells.
