Analysis of Modified Nucleotide Aptamer Library Generated by Thermophilic DNA Polymerases.

To select aptamers with modified nucleotides, the appropriate DNA polymerase must be designated. We studied eight polymerases and demonstrated that incorporating a tryptophan‐like nucleotide into aptamer libraries by emulsion PCR and KOD XL had an insignificant effect on the PCR bias and did not distort the sequence space, thus making this approach the method of choice.
