Genetic population structure of black grouper (Mycteroperca bonaci) in the northern coast of Yucatan

The structure of the black grouper (Mycteroperca bonaci) population from the northern coast of the Yucatan Peninsula was analyzed by means of genetic similarities among three separated (155–250 km) locations: Celestun, Dzilam, and Alacranes Reef. A total of 150 specimens were genetically characterized by means of five microsatellite DNA markers of the genome of this species amplified by PCR specific loci. Microsatellite amplicons were analyzed for each locality with subsequent genotyping in polyacrylamide gels. Results indicated three genetically distinct subpopulations (Celestun, Dzilam, and Alacranes Reef) that deviated significantly from Hardy–Weinberg equilibrium, and genetic differences were evidenced by the AMOVA, the pairwise genetic distance analyses, the Bayesian clustering analysis and the Principal Coordinate Analysis (PCoA). Thus, the analyzed samples do not belong to a black grouper panmictic population; instead they might represent a structured population along the northern coast of the Yucatan Peninsula that maybe caused by oceanographic-driven low levels of larval dispersal and gene flow between regions.
