Facile and Efficient Synthesis of Tri‐ and Tetrasubstituted 7‐Azabenzofuran Derivatives

A facile synthetic protocol for a series of tri‐ and tetrasubstituted 7‐azabenzofuran derivatives has been designed and successfully executed. The usefulness of this protocol is exemplified by the efficient synthesis of a potent HCV NS5B polymerase inhibitor. Compared to the known synthesis, the overall yield has been increased by six times. Those harmful and cost‐effective synthetic steps have been decreased, including reactions requiring the use of halogenating reagents, precious metals, high temperature or high‐pressure operation.
