Bioorthogonal metabolism ? Synthetic NAD analogue NCD has already been used to create bioorthogonal redox systems. We have engineered L. helveticus‐derived NAD‐dependent d ‐lactate dehydrogenase (Lh DLDH) to favor NCD by semirational design. Libraries were constructed from Lh DLDH‐V152R led to mutants with up to 940‐fold increased preference for NCD.
