The late stage S‐lipidation of unprotected peptides, a process we have coined CLipPA (Cysteine Lipidation of a Peptide or Amino acid) was optimized. The offensively smelly tBuSH was replaced with the more benign, t‐NonylSH and the lipopeptides could readily be recovered with an improved isolation procedure.
