Vinylboronic acids (VBAs) are stable, unstrained bioorthogonal reactants in tetrazine ligation. Fast reaction rates are obtained for tetrazines containing a coordinating substituent, whereas VBAs lacking a Lewis basic substituent are unreactive. We explored the molecular basis of the unique reactivity of VBAs and demonstrate the scope in the simultaneous orthogonal tetrazine labelling of two proteins.
