Tag team: 5‐Methylcytidine (m⁵C) and 5‐methyluridine (m⁵U) are highly abundant post‐transcriptionally modified nucleotides observed in various natural RNAs. These nucleotides are tagged through structure‐sensitive osmium complex formation. These complexes can be identified by mass spectrometry, which allows m⁵C and m⁵U to be distinguished from their isomers, 2′‐O‐methylcytidine and 2′‐O‐methyluridine, respectively.
