In the loop: We evaluated substitutions of two widely explored classes of cyclic and acyclic β‐amino acid into three different β‐hairpin loop prototypes within a β‐sheet‐rich protein tertiary structure, and identified sequence–structure– stability relationships that should be broadly extensible to future protein design efforts involving these residues.
