Testing the limits of modification: Nucleosides containing adducts at the O4 atom of 5‐fluoro‐2′‐deoxyuridine have been synthesised and incorporated into oligonucleotides by solid‐phase synthesis. The C5‐fluorine‐modified nucleosides display enhanced susceptibility towards removal of the O4‐alkyl group by O6‐alkylguanine DNA alkyltransferases (AGTs).
