Taking on water: Quenching of the final carbocation during terpene synthase catalysis, through proton loss or nucleophilic capture by water, is mediated by coordinated loop movements in the active site. A single amino acid switch in δ-cadinene synthase reduces hydrophobic contacts to allow water ingress during product release, resulting in an alcohol product.
