Crosslinks and conjugation: O6-Alkylguanine DNA alkyltransferases (AGTs) can act on O6-alkyl 2′-deoxyguanosine and O4-alkyl thymidine DNA lesions. We show the ability of the human AGT to efficiently repair intrastrand crosslinks, which can be used to generate covalent complexes. We synthesized IaCL DNA with ethylene or propylene linkages, with the latter being moderately repaired by human AGT.
