Inteins carry out protein-splicing reactions and can be used for genetic encoding and expression of cyclic peptides (CPs). We show that the target sequence and the DnaE intein strongly affect the intracellular peptide concentrations. The crystal structures of the intein pre-splicing complexes provide further molecular details on DnaE-mediated protein splicing.
