Siting cytidine: 2′-O-Methyl-5-hydroxymethylcytidine (hm⁵Cm), 5-hydroxymethylcytidine (hm⁵C) and 5-formylcytidine (f⁵C) phosphoramidite monomers have been synthesised and incorporated into RNA oligonucleotides. Optimisation of the mild post-synthetic deprotection conditions enabled the preparation of RNA oligomers containing several of one of the cytosine modifications or a combination of four.
