Diep et al. identify the molecular machinery of the yeast vacuole lipid droplet contact site vCLIP, which comprises two Vac8-dependent tethers, Ldo16 and Ldo45, and the phosphatidylinositol transfer protein Pdr16. vCLIP abundance and molecular composition are metabolically controlled, and vCLIP-dependent vacuole tethering is required for selective lipid droplet autophagy (lipophagy).
