Site-specific recombination in bacteria is involved in diverse processes such as integration and excision of bacteriophage genomes and successful segregation of chromosomes. One aspect of our research is directed at understanding how programmed DNA rearrangements are used by pathogenic bacteria to control expression of genes that are involved in the infection process. We are currently studying site-specific recombination in the bacterium Bacteroides fragilis that is a normal constituent of the human gut microflora. B. fragilis is also an opportunistic pathogen and can cause serious infections, such as peritonitis, following rupture of the gastrointestinal tract. A better understanding of pathogenicity determinants will allow more effective treatment in the future. Site-specific recombination in B. fragilis also controls expression of target site recognition proteins within a Type I restriction-modification (R-M) system. We are using both B. fragilis and E. coli as models to understand how R-M systems are controlled so that they do not destroy the resident bacterial chromosome.
