We work to genetically dissect the role/s of Fibroblast Growth Factor (FGF)/ FGF receptor (FGFR) signaling in mammalian organogenesis and stem cell function. In the past we have elucidated some of these roles in the developing craniofacial sutures, axial skeleton, limbs, lungs, genitalia, submandibular glands and the nervous system. Our model organism and research tool is transgenic mice that carry loss- or gain-of-function mutations in FGFs, FGFRs or FGF signaling mediator genes, or allow their conditional deletion or over-expression. In particular, we have used FGFR1 and FGFR2 mutant mice that model two congenital craniosynostosis diseases: Pfeiffer and Apert syndrome. Our current interest is focused on postnatal and adult neurogenesis in the hypothalamus, with relevance to regulation of appetite and energy-expenditure. Our in vivo studies are complimented by in vitro model systems to delineate the role of FGF signaling in progenitor cell growth and differentiation.
