>Secondary degeneration of intact tissue after traumatic central nervous system (CNS) injury is a major problem with no current therapies to prevent long-term functional decline. Inflammation, including the activation of resident microglial cells, plays an important role in these secondary changes. This multidisciplinary project aims to unravel the relationship between neuroinflammation and remote Blood Brain Barrier breakdown following neurotrauma and integrates pioneering nanoparticle technology to enable simultaneous delivery of therapeutics and imaging agents to halt or limit inflammation and remote BBB breakdown. >Targeting four-stranded DNA conformations to enable axonal regeneration following neurotrauma. In response to severe tissue damage or inflammation after severe trauma, reactive astrogliosis can lead to glial scarring. CNS scars consist of two distinct layers, fibrotic and glial. The glial scar forms the boundary between the fibrotic scar and the normal parenchymal tissues. Reactive astrocytes display increased expression of extracellular matrix components (ECM), including chondroitin sulfate proteoglycans (CSPGs), dramatic up-regulation of cytoskeletal proteins (Glial Fibrillary Acidic Protein (GFAP) vimentin and nestin) and increased expression of transcriptional regulators. As CSPGs have been implicated as barriers to CNS axon extension and are extremely inhibitory to axon outgrowth, the glial scar presents an obstacle for axonal regeneration. We are studying the implications of G-quadruplex DNA secondary structures in astrogliosis. It is well known that G4-DNA are implicated in transcriptional regulation. We are developing techniques to regulate formation of these structures in specific stress-induced genes in order to inhibit astrocyte activation and promote a regenerative environment >Targeting four-stranded DNA conformations to modulate gene expression in cancer and fibrosis.
