A multichannel closed bipolar electrode-LED sensing array for simultaneous sensitive and visible detection of multiple tumor markers

A novel closed bipolar electrode (cBPE) based click-light sensing strategy is prepared with integration of Au@MnO2 nanozyme for multi-mode visible detection of multiple targets, which have been widely applied in clinical tumor screening tests. cBPE benefiting from its easy requirement (one direct current power), portability, fast response and low background, has been an emerging device for construction of biosensors by integrating ultra-highly sensitive electrochemiluminescence (ECL) technique and shows great potential for point-of-care detection. However, existing cBPE-ECL methods are utilized at a high potential for ECL response, which greatly limits its sustainable and portable applications. Herein, a solid-state luminescent semiconductor, light emitting diode (LED) was first integrated with cBPE systems to provide significantly higher luminous efficiency in lower voltage and visualized fast readout in the sensing system. As target analytes (alpha-fetoprotein, prostate-specific antigen, and carcinoembryonic antigen) bind to the antibody-modified electrode surface, the nanozyme-labeled secondary antibodies (Ab2@Au@MnO₂) accelerate H2O2 decomposition, releasing more electrons. These electrons are transferred to the LED through the cBPE, increasing its luminous intensity, thus achieving simultaneous detection of multiple targets with low detection limits. Given the wide and urgent requirement of point-of-care devices, its potential applications in the construction of clinical sensing platform are expected.
