This review addresses the urgent need for more effective treatments for central nervous system (CNS) diseases, which currently affect one in six people worldwide. Despite advances in understanding their molecular and cellular mechanisms, CNS disorders remain difficult to treat due to their complexity and the protective nature of the blood-brain barrier (BBB).

The article highlights how multifunctional nanomaterials are emerging as promising tools to overcome these challenges. These platforms offer improved pharmacokinetics, targeted brain delivery, and the ability to modulate neuroimmune responses—making them particularly well-suited to tackle the multifactorial nature of CNS disorders.

We discuss key mechanisms underlying therapeutic resistance, recent innovations in nanoplatform design, and future directions for the rational development of multifunctional nanomaterials for CNS disease therapy.
