Development and characterization of 101 SNP markers in jumbo flying squid, Dosidicus gigas

Abstract

Dosidicus gigas (D. gigas) is a commercially important squid species in the Eastern Pacific Ocean. Due to the wide distribution of the species and its variable biological characteristics, there is a lack of reliable information regarding its genetic structure, which restricts the stock assessment and management of the D. gigas fishery resource. Therefore, there is an urgent need to develop sufficient molecular markers for the conservation and management of this species. In this study, 101 novel single nucleotide polymorphism (SNP) markers were developed using genotyping-by-sequencing (GBS). The observed heterozygosity (Ho) and expected heterozygosity (He) ranged from 0.0000 to 1.0000 and 0.0340 to 0.5000, respectively. The polymorphism information content ranged from 0.0333 to 0.3750. Twenty-five loci significantly deviated from the Hardy–Weinberg equilibrium after a Bonferroni correction (p < 0.001). These polymorphic SNPs will be useful for further population genetic analyses for D. gigas.
