Targeting TGFβ signalling in T helper cells

TGFβ1 produced by activated CD4+ T cells has been reported to suppress anti-tumour immunity, implicating an autocrine TGFβ T helper cell control circuit in cancer immune evasion. Here, Li et al. demonstrate that ablation of the TGFβ receptor type 2 (TGFBR2) in CD4+ T cells inhibits tumour growth in a mouse model of breast cancer resistant to immune checkpoint or anti-VEGF therapies. Furthermore, a bispecific receptor decoy, comprised of a TGFβ-neutralizing TGFBR2 extracellular domain attached to the non-immunosuppressive CD4 antibody ibalizumab, repressed TGFβ signalling in lymph node CD4+ T cells and reprogrammed the tumour vasculature, causing cancer cell hypoxia and cancer cell death in the mouse model.
