Abstract

A phytochemical investigation of Anemone vitifolia (Buch.-Ham.) led to the isolation of twelve compounds, including six lignans (16), four benzoic acid esters (710), one norsesquiterpenoid (11) and one lactone (12). The structures of these metabolites were established on the basis of detailed spectroscopic analysis, as well as comparisons with the data available in the literature. Among them, compounds 10 (dimethyl 2-(benzoyloxy)succinate) and 12 (4-hydroxy-5-methyl-γ-butyrolactone) were isolated for the first time as natural products. Compounds 1 and 11, compounds 2, 3, 57 and 9, and compounds 4 and 8 were isolated for the first time in family Ranunculaceae, genus Anemone and A. vitifolia respectively. The chemotaxonomic significance of the isolated compounds was discussed.

Subject and source

There are more than 150 species across the world in genus Anemone (Ranunculaceae), over 52 species are mainly distributed in Northeast, Northwest, and Southwest China (Deng et al., 2019). Anemone vitifolia Buch.-Ham. is mainly found in southwestern China, northern India, Sikkim, Nepal, northern Burma and Bhutan. And it was used as traditional Chinese medicine and was reported to be a potent analgesic, anti-inflammatory and antidiarrheal agent (Zhao et al., 2019; Bai et al., 2017).

A diverse range of compounds have been identified through chemical studies on the genus Anemone. The most common structure classes include triterpenoid saponins (Kwak et al., 2019; Anh Minh et al., 2012; Ding et al., 2011), lignans (Liu et al., 2020), flavonoids (Ganchimeg et al., 2019), norsesquiterpenoids (Thi Thanh Huong et al., 2019), alkaloids, lactones, volatile oils, and organic acids (Deng et al., 2019). Present study

The air-dried and powdered whole herb of A. vitifolia (6.5 kg) was extracted nine times with 95% ethanol (9 × 15.0 L, 1 h each) at room temperature. The obtained ethanol extracts were combined and concentrated under vacuum to give a dried extract (1.1 kg). The dried extracted was suspended in water and partitioned with petroleum ether, ethyl acetate, and n-butanol.

After removal of the solvent in vacuum, the ethyl acetate fraction (556 g) was separated through positive silica gel column

Chemotaxonomic significance

In this study, twelve compounds were isolated from the whole herb of A. vitifolia and elucidated, including six lignans (16), four benzoic acid esters (710), one norsesquiterpenoid (11) and one lactone (12). Compounds 10 and 12 were isolated for the first time as natural products, and they were obtained by chemical synthesis before.
